It utilizes a pump to force a liquid mobile phase through a column packed with a stationary phase under high pressure, allowing for rapid and efficient separation. The separated components are then detected and analyzed.
A High-Performance Liquid Chromatography (HPLC) instrument utilizes several key components to achieve sample separation. The Solvent Reservoir holds the mobile phase, the liquid that carries the sample through the system. A Pump forces the mobile phase through the system, ensuring consistent flow and pressure. An Injector introduces the sample into the mobile phase stream, typically using a syringe or autosampler. The Column contains the stationary phase, a material that interacts with the sample components, causing them to separate. The Detector monitors the separated components as they elute from the column, providing a signal that is then translated into a chromatogram by a Recorder/Data System. A Waste Reservoir safely collects the mobile phase and eluate after it passes through the detector. The entire system is designed to be automated, allowing for efficient and reproducible analysis of complex samples.
